Sports analogies are often overplayed, but it?s hard to resist thinking about football when discussing measures of success for strategic and business plans. What would the Super Bowl be like without first-down markers and a scoreboard? For all involved, the game would be almost meaningless.
Like football, business revolves around plans, people, action, and various ways to measure success. Statistics, metrics and measures tell the story. It?s now the end of the first calendar quarter. How do you and your team measure up to expectations? Yes, those expectations you set for yourself, and your team, when you created your strategic plan or business plan. What?s working and not working? Do you need to adapt and change course?
For some, this will be a friendly reminder. For those of you now ransacking your office to find a copy of your plan, this should be a wakeup call. Find your plan and follow along!
Company goals and objectives: While typically measured over a 12 month period, don?t wait until the end of the year to see how you?re doing. By then, it could be too late. Look to measure progress. For example, if one goal is to increase market share this year, you should be well on your way. Is your progress YTD acceptable?
Company performance indicators: This is your company?s dashboard: specific measures you should track regularly. They fall into one of four categories ? Customer Satisfaction, Organizational Culture, Internal Operations, and Financial Performance. Examples include revenue, profit, on-time delivery, customer acquisition, and employee retention.
Department goals and objectives: Similar to company goals and objectives, departmental goals will also include improvement initiatives. Using the market share example, suppose reaching your goal requires increased manufacturing efficiency or a new product line ?responsible departments should be tracking and reporting their progress.
Department performance indicators: Every department needs to perform and do their part. Measuring departmental performance drives accountability and improves the organization?s value stream ? cross functional performance.
Employee performance: Employees need to be growing professionally ? that is, if you want them to keep them. This goes beyond meeting basic? job specifications. It?s about talent management.
Everyone on a football team ?knows the score.? There are no secrets about what?s being measured. Consistency and transparency are critical for measuring business performance too. Make sure your team knows what?s being measured and how success is defined and your accomplishments will grow quarter by quarter.

We?ve read mixed reports about how lofty gold and silver prices are affecting demand in India. One month we?re told demand is up, and the next it?s supposedly down. I?m not suggesting that official reports are inaccurate, but it is admittedly confusing and doesn?t help us understand the real trend in the country.
Why should we care about the gold market in India? Well, let?s face it; the nation is one of the biggest consumers of the metal, a major driver that can give us hints about demand and investment trends, along with what to perhaps eventually expect here in North America. But reading third-party reports about India is very different than getting information firsthand from a credible source in the country. I wanted to get to the bottom of what?s really going on in India by talking to a reputable bullion dealer who could give me the inside scoop, an up-to-the-moment dispatch from the front lines, as it were. So I did just that.
Ashish and Rashmi Sand own Savio Jewellery (Savio means ?shine? in Italian), a design studio and jewelry factory in Jaipur, India. They?ve received many design and manufacturing awards since starting their business six years ago, winning five awards in just the past six months. They source gold from bullion agents in Jaipur, who in turn obtain it from dealers in Hong Kong, Dubai, Mumbai, and Delhi. They have industry contacts, friends, and relatives that span the globe, from the US and UK to Asia and Australia. If anybody knows what?s happening in the physical gold and silver bullion markets and the Indian jewelry market, it?s them.
In this exclusive interview, you can read what Ashish and Rashmi told me about unstoppable demand, growing silver interest, budding demand for coins and bars, reduced selling, shifting trends with women, burgeoning ETFs, and why they believe a bubble is headed our way?
Jeff Clark: Ashish, tell us about your manufacturing facility and design studio.
Ashish: Savio Jewellery makes conceptual jewelry, which can be worn in five or six different ways. It looks ?heavy? but is extremely lightweight. It?s also affordable.
We design what you want, for as much as you want to spend. For instance, one of my customers wanted a necklace and earring set for her daughter?s wedding; based on their budget, we were able to manufacture a set using 35 grams of gold [about 1.2 ounces].
Jeff: Do you have any difficulties sourcing metal?
Ashish: We don?t face any difficulties in arranging gold, whether demand is high or low, because the traders hedge gold on daily basis. We also keep a reserve of gold so that our manufacturing doesn?t slow down if metal isn?t immediately available. Our suppliers of gold don?t face any problem procuring gold, either.
Jeff: How would you describe demand for gold right now in India?
Ashish: We are seeing several shifts in the trends for gold demand. First, even with the increase in price, demand is still strong. People have not stopped buying. Part of this is cultural; wearing gold jewelry in a wedding is a longstanding custom in India and will not change regardless of price.
In fact, our shops are experiencing increased buying in spite of higher prices. Previously we would have one or two customers per day for gold, but now we have five or six. We are getting plenty of new customers, and see new customers more frequently now.
Second, I would say that awareness among customers has increased. Now they see jewelry also as an investment and want a proper return on their investment. Further, due to fluctuations in price, they may book in advance ? i.e., if a customer orders a jewelry product, they lock in the gold price ahead of time.
Probably the biggest change we face, though, is that customers are more focused on getting lightweight jewelry, or 14-carat gold instead of 18-carat gold. This is due to the high gold price. This adjustment, however, has not affected the desire to own gold.
The bottom line is that the rise in the gold price has not hampered demand.
Jeff: Are Indians investing more in bullion (bars and coins) than usual?
Ashish: Indians are definitely investing more in bullion due to the rising trend in gold and silver prices. Investing in gold and silver has always been a good idea for wealth protection to Indians, and now we are seeing them buy more coins and bars than they used to. Coins and bars are mainly purchased either in wedding season or during the Diwali festival. Even I purchase gold coins every Diwali ? 10 grams, 20 grams, or any amount.
Another factor is that India has experienced many monsoons over the last two years, and the farmers are tending to invest their savings in gold and silver. This is a major factor for the increase in demand for coins and bars.
Jeff: Is much reselling taking place right now? Is it higher or lower than it used to be?
Ashish: Reselling gold has become rare ? and this from a country that consumes more gold than any other and where gold jewelry is a central part of the culture. We rarely have customers who sell back their gold or silver. Before we used to have customers who would sell their scrap gold and make new jewelry from it, but now we don?t experience many such customers. I would estimate we currently have 90% buyers and 10% sellers, and before the ratio was 80% buyers and 20% sellers.
They?re not selling, because they hope that in the next few years the price will rise further and they can get better returns than what they can at present. Just a few years ago, some customers would buy gold expecting to sell when the price rose; now those customers are sure gold will keep rising year after year and are selling only rarely.
Jeff: We?ve read about an increased interest in silver in India ? tell us what you see.
Ashish: Silver demand has increased sharply in recent years, as increasing numbers of investors use silver as a store of value. Customers are also buying silver to hedge against market losses. While global markets have been uncertain over the past four years, silver has provided huge returns and has been an ideal investment. Because silver has provided the highest recent return of all the commodities, this has attracted the average layman.
Silver demand has also increased for the customer for whom the gold price is out of reach. Nowadays, if they cannot afford gold, they switch to silver. So the number of customers requesting silver has increased.
Silver is also part of gift-giving in India. It is customary to give silver items as a wedding gift, and there are rituals where parents gift silver dinner sets to their daughters at their weddings. In addition to dinner sets, silver is usually found in jewelry, such as a necklace, earring, bracelet, or rings; gift items such as glass sets and crafted boxes; and even showpiece furniture.
Last, there is a steep rise in demand for silver in electronic goods.
Jeff: Rashmi, describe how Indian women view gold. And have you noticed any changes in how they view silver?
Rashmi: Gold and silver are firmly embedded in cultural and religious traditions for Indian women. Over the past decade, Diwali and Akshaya Tritiya have become major gold- and silver-buying occasions in India. Gold and silver also play an important role in the marriage ceremony, where brides are often adorned from head to toe in gold jewelry. This reminds me of my own wedding.
[Rashmi is wearing approximately 500 grams of pure gold (about 17.6 ounces), including a necklace, earring, a tika and matha patti on her head, haath phool in her hands, bangles on her wrists, nose ring, arm band (bajubandh), and finger rings.]
So during wedding season, the demand for gold and silver increases. Our women customers prefer lightweight jewelry instead of heavy and bulky ornaments. Increasingly, it also appears they now prefer workmanship over weight when choosing jewelry made from gold. The Indian custom of women wearing jewelry has not changed, though they have started accepting 14 carat gold in order to make it more pocketbook friendly. And people in India wear jewelry to all kinds of parties.
We have also experienced an increased number of women customers in our shop. They still buy gold and silver jewelry, but now they prefer jewelry that is of designer quality and looks good. This is the biggest change I have noticed with women over the past few years; they want jewelry that is unique. They prefer wearing long earrings ? danglers or hoops.
They also prefer multipurpose or conceptual jewelry ? in other words, jewelry that can be worn in different ways. This attracts the woman customer, as they think they have more jewelry for the same price.
There is a different culture in south India, where the people are more bound to gold jewelry than silver. The parents still focus on giving gold to their daughters for a wedding present. Each part of India has different customs, but the one thing that remains common is that gold jewelry is bought by everyone for weddings.
Jeff: Ashish, you?ve stated you are very bullish on the price of gold and silver, and that a bubble for prices is ahead; why?
Ashish: To be clear, we are extremely bullish on the price of silver and gold. We believe that both will eventually advance into a full-fledged bubble that will surpass most investors? wildest dreams.
The world is changing and so is the way we invest. Gold and silver were once considered a marginal investment, worn mostly as jewelry or something that was stored in banks. Now it is making a comeback and has become a primary investment asset for many individuals and institutions.
In India, people are very futuristic and are so bound to the culture that they?re buying gold and silver in advance. If they know that they are going to marry their daughter in three to four years, for example, they will purchase the gold now instead of waiting when prices will be higher. This will continue to underpin demand and push the price higher.
Jeff: Any other trends you see in India?
Ashish: From the average layman to the high-class educated person, they now want to invest in gold and silver. In fact, those who were investing in equities have been switching to gold and silver. Even the gold and silver ETF has become common in India. This is partly due to rising prices; keeping metal at home has become riskier, so they can avoid the risks of self-storage by using an ETF.
Even I have invested in gold and silver for my personal use apart from my business. Instead of buying equities, I now invest more in gold and silver, with the expectation of getting a good return.
I will conclude by just saying that based on what we see here every day, we believe gold and silver prices will continue to go up.
Jeff: Thanks, Ashish and Rashmi.

Large organizations and companies need to have constant and real time communication in order for them to facilitate their transactions and other operations. The various departments of an organization must have smooth communication and transmissions. For that purpose, proper voice and data cabling is very important. This must be installed in the network or communication systems.
This would be necessary if a company is adding one work station or a new network. If an organization plans to upgrade their existing system or wants to have a whole new one installed, this is also perfect for those needs. A fully functioning system would benefit any group or organization especially in the world of business.
Failure in communication can affect the operations of any organization. This may manifest in several ways such as downtime, slow response time and faulty transmissions. These instances can cause delays in operations and other important functions of a company. If this is not resolved sooner, this would lead to losses in the business or even misunderstanding.
To avoid worse case scenarios, a well designed system must be installed. This type of system must be correctly configured to make sure everything in the system is working properly. Prior to configuration or installation, it must start with the design. This must be done in way that it can provide the specific needs of a company or organization.
Different types of wiring can be used to build this network. These may include shielded copper, multi pair copper, fiber optics and other wiring mediums. Cables used in this system are typically color coded to indicate the type of connection they have. It will be easier for technicians to identify the wires in case of any problem along the way.
When installing a network, it is best to make sure that the system is fault free. Again, this will depend on the design, configuration and proper installation. Making sure that everything is working well can save time and money from long term troubleshooting when the network encounters technical problems. Everything must be thoroughly checked before starting the operation.
In this case, organizations that need a system to be installed should look for the right team of professionals. The team must have engineering qualifications, technical knowledge and expertise in design. Experience in installation is also important. The success of this job relies a lot on the efficiency and expertise of professional installers.
Organizations should get the best in the field to ensure efficiency and optimal function of their networks and systems. This complex task cannot be done by meager providers. This job can only be done by those who actually have experience and are well rounded in this matter. After all, this involves wiring and various connections.
When it comes to voice and data cabling, professional installers also follow standards which are set by proper associations or boards in this field. These standards are made for the purpose of safety and proper function. These are already used internationally and even published for installers to follow when they provide services to their clients.

MixStirs franchising began in 2007 offering smoothies ? or ?Stirs? as the company prefers to call them ? for active lifestyles on the go. MixStirs franchises are primarily found in small locations with captured audience venues such as fitness centers, kiosks, malls, transportation centers and sporting arenas with a few full-service stores.
The company?s president, Mike Greenberg, says MixStirs franchises can be built in a wide variety of space configurations but ideal locations for MixStirs are typically 400 to 800 square feet. ?We focus on small locations to help limit startup costs and overhead making it much easier for our franchisees to be profitable and successful,? explained Greenberg.
Greenberg says MixStirs has yet to have a store close because of poor performance which he credits to the low overhead involved in the operation, and that the company would rather open five to 10 healthy stores per year than 25 and have five stores close.
According to Greenberg, MixStirs franchising success can also be attributed to the level of support the company offers its franchisees. ?At MixStirs we pride ourselves with our support. We support our franchisees in all aspects of their business and none of them will tell you different. Our support ranges from helping an individual location develop a marketing plan to helping them hang up a sign. Whatever our franchisees need we support them with,? said Greenberg.
Greenberg believes that the smoothie market still has a lot of potential to grow as Americans continue to lean towards healthier menu choices. He says that although most people have heard of a smoothie, the beverage is still very low as a percentage of food and beverage sales. ?The requirement of calorie information on many menu boards is starting to help people to make healthier food choices and will continue to move healthy fruit smoothies, like the ones served at MixStirs, up the ladder.?
Good candidates for a MixStirs franchise should be open minded and possess a strong willingness to learn the franchise system according to Greenberg who believes that anyone can make a smoothie, but that franchisees need to learn MixStirs? specific way of making smoothies in order to deliver the best possible product to customers.
Franchise fees for MixStirs cover the use of the company?s trademarked logo and recipes as well as all training and support to get the location open which includes both off and on-site pre-opening training. Royalty fees are used for ongoing support, training, quality assurance, service and cleanliness evaluations, and more.
Currently, MixStirs has locations in six states and is offering smoothie franchises across the country.

ScienceDaily (Mar. 24, 2012) ? More than 50% of the world's plant species have been discovered by 2% of plant collectors, scientists have found.
With an estimated 15-30% of the world's flowering plants yet to be discovered, finding and recording new plant species is vital to our understanding of global biodiversity.
The age of great botanical explorers, such as Sir Joseph Banks and Alexander von Humboldt, might appear to have passed. But the study, led by Oxford University scientists, found that modern botany has its own 'great plant hunters' -- individuals whose experience and skills enable them to make a disproportionate contribution to the discovery of new plant species.
A report of the research was recently published in the Proceedings of the Royal Society B.
'It seems that, even in the 21st Century, we need 'great' plant hunters who have the skills and experience to make the most efficient use of their time in the field,' said Dr Robert Scotland of Oxford University's Department of Plant Sciences, who led the work.
'Whilst local specialists, citizen scientists, and students all have an invaluable contribution to make to botany, our research suggests that years of experience helps great hunters collect, not necessarily more specimens, but more of the important ones that go on to change our understanding of plant species,' Dr Scotland adds.
The study assembled four datasets totalling 100,000 specimens from four institutions; The Natural History Museum, Royal Botanic Garden Edinburgh, Missouri Botanical Garden, and Royal Botanic Garden Melbourne.
The researchers found that the most productive collectors are distinguished by five attributes: they collect over many years, they collect more types per year, they collect from several different countries (although specialising in one particular country), they collect from a wide range of plant families (although again, often specialising in a particular family), and they collect more types towards the end of their careers.
The study suggests that greater efforts should be made to identify, train, and support plant hunters throughout their careers as they can make a substantial contribution to the discovery of new species.
Oxford University's Department of Plant Sciences has a strong history of producing 'big hitting' plant hunters from an 18th Century Professor of Botany, John Sibthorp (1758-96) who collected some of the first plants from Greece and many from Cyprus, to Sir Ghillean Prance, a recent Director of Kew Gardens and Graduate student at Oxford University who collected extensively in Brazil. Currently John Wood, a research associate at the Department of Plant Sciences, has collected 30,000 specimens from South America and Asia, many of which are new species.

The Pentagon searches for technology to treat tinnitus, characterized by a persistent ringing in the ears, and suffered by one in 10 U.S. adults, and 40 percent of all veterans

HEAR THAT?: Draper Laboratory in Cambridge, Mass., is fleshing out a concept for a small delivery device that can provide relief to tinnitus sufferers. The device would be inserted near the membrane-covered window separating the middle ear from the inner ear and release medication into the cochlea.Image: Courtesy of Draper Laboratory
Loud, concussive explosions on the battlefield may last only a few seconds, but many soldiers returning from combat in the Middle East are experiencing lingering symptoms that cause them to perceive sounds even when it is quiet. Doctors can do little to treat the problem?typically described as a ringing in the ears?because they lack an effective way of delivering medication to the inner ear. That could change in a few years, in the form of an implantable polymer-based microscale drug-release system that delivers medicine to the inner ear.
Called tinnitus, the condition afflicts at least one in every 10 American adults and is the most common disability among Afghanistan and Iraq war veterans, according to the U.S. Department of Veterans Affairs (VA). Up to 40 percent of all veterans may be suffering from tinnitus, and the VA spends about $1 billion annually on disability payments for tinnitus, according to a study published last year in Nature. (Scientific American is part of Nature Publishing Group.)
To address the problem, the U.S. Department of Defense has commissioned Draper Laboratory in Cambridge, Mass., to spend the next year fleshing out a concept for a small delivery device inserted near the membrane-covered window?no more than three millimeters in diameter?separating the middle ear from the inner ear. Once at the membrane the device (essentially a polymer capsule, although Draper is not developing any of medicines that might be placed inside) would release a drug into the cochlea, the tubular organ residing in the inner ear that enables us to hear. The plan is to embed wireless communications into the capsule so that a patient or doctor can control the dosage. After the capsule finishes delivering its supply of drugs, it would dissolve.
Although Draper's project is still in the very early stages and years away from any clinical testing, it holds more promise than many of today's most common approaches to tinnitus treatment, which include deep breathing, using background noise to drown out the ringing or simply learning to ignore the bothersome sound. Steroids injected into the eardrum have shown some promise in helping patients with certain hearing and balance disorders, but the ear begins eliminating these drugs through the eustachian tube (a passageway in the middle ear that acts as a pressure equalizer) as soon as the patient talks, swallows or even sits up. As a result, the patient must endure several injections into their ear and remain immobilized for a time after each injection to get any relief from the malady.
"By and large there aren't that many good ways to treat tinnitus," says Lloyd Minor, provost and senior vice president for academic affairs at Johns Hopkins University. Draper's work "is potentially a novel way of delivering drugs to treat tinnitus. In general, we don't have the types of drug-delivery systems that we would like to get medication into the inner ear."
NeuroSystec Corp. is developing a neuro-active agent designed to calm the hyperactive nerves responsible for cochlear tinnitus. The Valencia, Calif., biotech start-up has licensed an osmotic pump from Durect Corp. in Cupertino, Calif., a company working on a number of drug delivery mechanisms for various parts of the body, including the inner ear.
Other advanced approaches of addressing tinnitus have been in the works for years, but most are still not ready for the market. Otonomy, Inc., in San Diego is testing a sustained release dexamethasone (a type of steroid) gel that would be injected into the middle ear, where it would stay in place, dissolving slowly and delivering treatments for hearing and balance disorders. MicroTransponder, a medical device company spun out from the University of Texas at Dallas in 2007, is looking to broaden the use of its implanted wired neuro-stimulation system for treating epilepsy to likewise help tinnitus patients.
The neuro-stimulation approach shows greater promise than those based on delivering medication to the inner ear at this time, says Michael McKenna, an otologist and neurologist at Massachusetts Eye and Ear in Boston. Targeted drug therapy is of questionable benefit because tinnitus comes from a variety of causes?including age-related hearing loss, traumatic ear injuries or circulatory system disorders?and has varying degrees of severity, he adds.
Perhaps some combination of all these efforts will end up delivering the relief that tinnitus sufferers seek. "Nothing really has been a panacea, so there is the need for further technological development," Minor says. If Draper's technology "works in the way they're hoping it will work, it will potentially be a big advance for the field."

Michigan State's surprisingly successful season ended with a thud.
The Spartans started the season unranked, then accomplished enough to be a top-seeded team in the NCAA tournament. They advanced to the round of 16 for the 10th time in 15 years, but ended with perhaps their poorest performance of the season.
Louisville sent Michigan State home with a 57-44 win Thursday night in the West Regional semifinals.
"It's hard to feel good and hard to feel bad," Spartans coach Tom Izzo said. "It's hard to feel good because I don't think we had anybody that played at all like they played in the last three weeks. But it's hard to feel bad because Louisville deserved it."
Michigan State entered the game on a roll, winning three games to earn the Big Ten tournament title and its first two games in the NCAA tournament.
The Spartans saved their worst for last. They made just 29 percent of their shots overall and got outrebounded by the Cardinals, whose zone defense slowed down a previously effective offense.
"We thought we was pretty well prepared for it," forward Draymond Green said. "It's not our coaches' fault. I think they gave us a great game plan. At the end of the day, players play, and we didn't, we just didn't execute well."
Green, though, did for much of the season.
The do-it-all forward was chosen the Big Ten player of the year, the conference tournament's most outstanding player and broke Greg Kelser's school record for career rebounds.
Green is graduating this spring, leaving the program with a big void on and off the court. The Spartans also will be without departing seniors Austin Thornton and Brandon Wood.
Michigan State returns a solid nucleus of players, including point guard Keith Appling, centers Derrick Nix and Adreian Payne along with guard Branden Dawson, whose freshman season was cut short by a left knee injury. Dawson has had surgery and hopes to be ready to join the team for preseason practices next fall.
The Spartans have a highly touted recruiting class, including two of the top in-state players, Bay City's Matt Costello, who won the Mr. Basketball award and runner-up Denzel Valentine of Lansing, along with Gary Harris, who was recognized as the top player in Indiana, and Ohio's Kenny Kaminski.
Michigan State won the Big Ten title for the third time in four years, claimed the conference tournament championship for the first time since 2000 and advanced to the NCAA's round of 16 for the 10th time in 15 years.
For the first time in four NCAA tournaments as a No. 1 seed, Izzo didn't guide the Spartans to at least four wins to reach the national semifinals. His record fell to 7-3 in the round of 16, and he wasn't happy about preparations for the first regional semifinal with just three days off.
The team flew to Arizona on Monday night, a day after beating Saint Louis, to prepare for Louisville.
"I think I made some mistakes, too, I really do," Izzo said. "I'd never come out on a Monday again. I think we needed more time (in East Lansing). It was just a weird situation for us.
Michigan State started the season in an unusual position, not ranked in The Associated Press preseason poll. The Spartans had just two players ? Green and Appling ? who averaged double digits in minutes last year.
After setbacks in showcase games against North Carolina on an aircraft carrier and to Duke in New York, the Spartans won 15 straight, including at Gonzaga, against Indiana and at Wisconsin. They then lost three of five in January and rallied with a seven-game winning streak that gave them a piece of the Big Ten title. They ended up sharing it by closing the regular season with two straight losses.
Izzo and his players overcame the loss of standout Dawson at the Big Ten tournament in wins over Ohio State, Wisconsin and Iowa and with two victories in the NCAA tournament.
"We had a special year," Thornton said. " We know we did some things at Michigan State that haven't been done in a long time
"Our ultimate goal was to make it to New Orleans to the Final Four. We weren't able to accomplish that."
Izzo said no one played well, or coached well, in the finale.
"We grew together, we won together, we had fun together, we cried together," he said. "You couldn't even look at one guy or blame one guy; collectively we just didn't get it done."

ScienceDaily (Mar. 21, 2012) ? Researchers at Weill Cornell Medical College have developed a computer program that has tracked the manner in which different forms of dementia spread within a human brain. They say their mathematical model can be used to predict where and approximately when an individual patient's brain will suffer from the spread, neuron to neuron, of "prion-like" toxic proteins -- a process they say underlies all forms of dementia.
Their findings, published in the March 22 issue of Neuron, could help patients and their families confirm a diagnosis of dementia and prepare in advance for future cognitive declines over time. In the future -- in an era where targeted drugs against dementia exist -- the program might also help physicians identify suitable brain targets for therapeutic intervention, says the study's lead researcher, Ashish Raj, Ph.D., an assistant professor of computer science in radiology at Weill Cornell Medical College.
"Think of it as a weather radar system, which shows you a video of weather patterns in your area over the next 48 hours," says Dr. Raj. "Our model, when applied to the baseline magnetic resonance imaging scan of an individual brain, can similarly produce a future map of degeneration in that person over the next few years or decades.
"This could allow neurologists to predict what the patient's neuroanatomic and associated cognitive state will be at any given point in the future. They could tell whether and when the patient will develop speech impediments, memory loss, behavioral peculiarities, and so on," he says. "Knowledge of what the future holds will allow patients to make informed choices regarding their lifestyle and therapeutic interventions.
"At some point we will gain the ability to target and improve the health of specific brain regions and nerve fiber tracts," Dr. Raj says. "At that point, a good prediction of a subject's future anatomic state can help identify promising target regions for this intervention. Early detection will be key to preventing and managing dementia."
Tracking the Flow of Proteins
The computational model, which Dr. Raj developed, is the latest, and one of the most significant, validations of the idea that dementia is caused by proteins that spread through the brain along networks of neurons. It extends findings that were widely reported in February that Alzheimer's disease starts in a particular brain region, but spreads further via misfolded, toxic "tau" proteins. Those studies, by researchers at Columbia University Medical Center and Massachusetts General Hospital, were conducted in mouse models and focused only on Alzheimer's disease.
In this study, Dr. Raj details how he developed the mathematical model of the flow of toxic proteins, and then demonstrates that it correctly predicted the patterns of degeneration that results in a number of different forms of dementia.
He says his model is predicated on the recent understanding that all known forms of dementia are accompanied by, and likely caused by, abnormal or "misfolded" proteins. Proteins have a defined shape, depending on their specific function -- but proteins that become misshapen can produce unwanted toxic effects. One example is tau, which is found in a misfolded state in the brains of both Alzheimer's patients and patients with frontal temporal dementia (FTD). Other proteins, such as TDP43 and ubiquitin, are also found in FTD, and alpha synuclein is found in Parkinson's disease.
These proteins are called "prion-like" because misfolded, or diseased, proteins induce the misfolding of other proteins they touch down a specific neuronal pathway. Prion diseases (such as mad cow disease) that involve transmission of misfolded proteins are thought to be infectious between people. "There is no evidence that Alzheimer's or other dementias are contagious in that way, which is why their transmission is called prion-like."
Simple Explanation for Clinically Observed Patterns of Dementia
Dr. Raj calls his model of trans-neuronal spread of misfolded proteins "very simple." It models the same process by which any gas diffuses in air, except that in the case of dementias the diffusion process occurs along connected neural fiber tracts in the brain.
"This is a common process by which any disease-causing protein can result in a variety of dementias," he says.
The model identifies the neural sub-networks in the brain into which misfolded proteins will collect before moving on to other brain areas that are connected by networks of neurons. In the process the proteins alter normal functioning of all brain areas they visit.
"What is new and really quite remarkable is the network diffusion model itself, which acts on the normal brain connectivity network and manages to reproduce many known aspects of whole brain disease patterns in dementias," Dr. Raj says. "This provides a very simple explanation for why different dementias appear to target specific areas of the brain."
In the study, he was able to match patterns from the diffusion model, which traced protein disbursal in a healthy brain, to the patterns of brain atrophy observed in patients with either Alzheimer's disease or FTD. This degeneration was measured using MRI and other tools that could quantify the amount of brain volume loss experienced in each region of the patient's brain. Co-author Amy Kuceyeski, Ph.D., a postdoctoral fellow who works with Dr. Raj, helped analyze brain volume measurements in the diseased brains.
"Our study demonstrates that such a spreading mechanism leads directly to the observed patterns of atrophy one sees in various dementias," Dr. Raj says. "While the classic patterns of dementia are well known, this is the first model to relate brain network properties to the patterns and explain them in a deterministic and predictive manner."
The study's third contributing author is Michael Weiner, M.D., professor of radiology at the University of California at San Francisco, and the principal investigator of the Alzheimer's Disease Neuroimaging Initiative, funded by the National Institutes of Health.

PHNOM PENH, Cambodia (AP) ? Myanmar's president confirmed Wednesday that his country will allow some foreign election observers, including from the United States and Europe, to monitor next month's polls, considered a crucial test of reforms he has taken in the military-dominated country.
Myanmar President Thein Sein, making an official visit to Cambodia, said each of the 10 member countries of the Association of Southeast Asian Nations would be allowed to send two official delegates to watch the April 1 by-elections, according to Cambodian government spokesman Khieu Kanharith.
Khieu Kanharith cited Thein Sein telling Cambodian Prime Minister Hun Sen that ASEAN's so-called dialogue partners ? Australia, Canada, China, the European Union, India, Japan, New Zealand, South Korea, Russia and the United States ? would also each be invited to send a small delegation, as would ASEAN's own administrative office.
It was not immediately clear what sort of access would be granted to the observers, but their invitation is considered a breakthrough. Myanmar's former ruling junta had rejected international bids to observe the last two elections, in 2010 and 1990.
The National League for Democracy, Myanmar's main opposition party led by Nobel peace laureate Aung San Suu Kyi, has expressed concern about the possibility of electoral fraud. The party solidly won the 1990 elections but was never allowed to take office, and it boycotted the 2010 elections as neither free nor fair. Its participation in the April vote is seen as an endorsement of the reforms Thein Sein has undertaken since taking office last year.
Khieu Kanharith said Thein Sein had asked Hun Sen for information on its experiences with foreign election observers and the Cambodian leader agreed he would send some documents on the subject. Hun Sen, like Thein Sen, has been accused of using the democratic process to front for authoritarian rule.
Thein Sein leads an army-backed party that won a landslide of seats in the 2010 polls. Although he has taken reform measures such as easing limits on the press and freeing political prisoners since taking office a year ago, he rules under a constitution that ensures that the military retains the dominant role in politics.
ASEAN had said Tuesday poll watchers from regional countries were invited, and the U.S. Embassy in Myanmar confirmed the expanded invitation Wednesday.
"We are encouraged that the authorities have invited international representatives as observers, including from ASEAN, the EU and the United States," embassy spokeswoman Adrienne Nutzman told The Associated Press. She said an invitation was sent to Washington.
"It's notable that the Burmese government will also allow some journalists from these countries to observe the process, demonstrating increased openness to the foreign media," Nutzman said. The U.S. government refers to Myanmar by its old name, Burma.
She declined to give details of how many American monitors would be allowed and if the U.S. had accepted the invitation, referring further questions to Washington.
The April by-election is being held to fill 48 parliamentary seats vacated over the past year.
Suu Kyi, a former political prisoner, is running for one of the seats in parliament and has generated enormous support for her party in a nationwide campaign tour. If Suu Kyi wins, she would have a voice in government for the first time after decades as the country's iconic opposition leader.
The United States and other countries have said they will wait to see if the by-elections are free and fair before they consider lifting political and economic sanctions imposed against the former military regime.
